You can turn the car in at a difffernt location than the one from where you picked it up. Negate the parking charges and get on your way. There's ZERO reason to have a car in Paris - and your stay won't be dictated by where you can find (and pay) for a parking!
Rather than renting a car at the start and going through the hassle of where to park it, why not rent it only on the days you really need to have it? I'm not familiar with La Defense, but some of the hotels I have stayed at charge a pretty stiff fee to park the car. If the rental is for going out of Paris, chose a rental pickup point at one of the Portes on the Peripherique. I personally use Porte Maillot rental pickups because you can easily pick up the Autoroute at La Defense for points west.

You can either house the car in a parking and pay the daily rate. There are many hotels located in La Denfense where parking costs little to nothing. If you select one of them, plan on commution by RER.
Many of these hotels are part of the Mecure Group.
